1 2 Alzheimer ' S Disease , Brain Injury , and C . N . S . Nanotherapy in 3 Humans : Sonoporation Augmenting Drug Targeting

ABSTRACT: 10 Owing to the complexity of neurodegenerative diseases, multiple cellular types need to be targeted 11 simultaneously in order for a given therapy to demonstrate any major effectiveness. Ultrasound12 sensitive coated microbubbles (in a targeted nanoemulsion) are available. Versatile small-molecule 13 drug(s) targeting multiple pathways of Alzheimer's disease pathogenesis are known. By 14 incorporating such drug(s) into the targeted LCM/ND lipid nanoemulsion type, one obtains a 15 multitasking combination therapeutic for translational medicine. This multitasking therapeutic 16 targets cell-surface scavenger receptors (mainly SR-BI), making possible for various Alzheimer's17 related cell types to be simultaneously searched out for localized drug treatment in vivo. Besides 18 targeting cell-surface SR-BI, the proposed LCM/ND-nanoemulsion combination therapeutic(s) 19 include a characteristic lipid-coated microbubble [LCM] subpopulation (i.e., a stable LCM 20 suspension); such LCM substantially reduce the acoustic power levels needed for accomplishing 21 temporary noninvasive (transcranial) ultrasound treatment, or sonoporation, if additionally desired 22 for the Alzheimer's patient. 23 24
